New Enzyme PecI — a True Isoschizomer of the Restriction Endonuclease PsiI

We have identified a bacterial strain, Paracoccus species 12, which is a producer of a novel restriction endonuclease designated PecI. The enzyme recognizes the palindromic hexanucleotide DNA sequence
5′-TTA↑TAA-3′ and cleaves it to generate blunt ends, as indicated by the arrow. Thus, PecI is a true isoschizomer of the restriction endonuclease PsiI.

Compared to PsiI, PecI offers several advantages, including higher enzyme concentration and 100% activity in ROSE buffer, which enables the availability of a Turbo version for rapid DNA digestion.
