Unit definitionOne unit of activity is defined as the amount of enzyme required to digest 1 µg of bacteriophage T7 DNA in 50 µl of reaction mixture containing SE-buffer W at 37°C for 1 hour.
Assayed onBacteriophage T7 DNA
Storage conditions10 mM Tris-HCl (pH 7.6), 200 mM NaCl, 0.1 mM EDTA, 7 mM 2-mercaptoethanol, 50% glycerol. Store at −20°C.
LigationAfter digestion with a 3-fold excess of enzyme, DNA fragments are poorly ligated by T4 DNA ligase. Ligation efficiency exceeding 95% is achieved in the presence of 10% PEG.
Nonspecific hydrolysisThe specific DNA digestion pattern remains unchanged after incubation of 1 µg DNA with 6 units of enzyme for 16 hours at 37°C. No nonspecific hydrolysis of single- or double-stranded oligonucleotides is observed after incubation with 3 units of enzyme for 3 hours.
