SibEnzyme announces the commercial availability of SegI, a new rare-cutting Type II restriction endonuclease isolated from Sphingobium erigeronicola 46.
SegI recognizes the degenerate palindromic octanucleotide sequence 5′-RTGC↓GCAY-3′, where R = A or G and Y = C or T, and cleaves both DNA strands at the center of the recognition site, generating blunt ends.
The enzyme is an isoschizomer of FspAI and provides an additional option for applications requiring the cleavage of DNA at specific 8-bp recognition sequences.
Key Features
- Recognition site: 5′-RTGC↓GCAY-3′
- Cleavage pattern: Blunt ends
- Isoschizomer: FspAI
- Optimal reaction temperature: 37°C
- Recommended reaction buffer: SE buffer W
- Enzyme activity: 5,000 U/mL
SegI is suitable for applications requiring large DNA fragments, including molecular cloning, DNA mapping, genetic engineering, and other molecular biology workflows.
SegI was developed and characterized by researchers at SibEnzyme. The results of this study, including the determination of the recognition sequence, cleavage position, and optimal reaction conditions, are presented in the following scientific publication: Gonchar D.A., Chernukhin V.A., Abdurashitov M.A., Dedkov V.S., Mikhnenkova N.A., Belichenko O.A., Degtyarev S.Kh. (2026). SegI restriction endonuclease from Sphingobium erigeronicola 46 recognizes the octanucleotide DNA sequence 5′-RTGC↓GCAY-3′ and is an isoschizomer of FspAI. DNA-Recognizing Enzymes, 2026(5).
SegI is now commercially available from SibEnzyme. Contact our team for ordering information and product details.
